When running on muffs how long will the engine have to run before water comes out the pee hole

I would start to worry if no water appeared within 15 seconds.

It depends if there was water in the system before hand by keeping the motor up.
It can take up to a couple of minutes depending on how much pressure you are feeding it at the hose.
After 2 minutes, I would shut it down.

I would say a minute or minute and a half. On the long side. I'm sure you will get a lot of different answers as people that answer you may be running anything from a 25 horse power to a 225 horse power. From 1960 year model to 2114 models. As long as you have water on the foot you know you are keeping the impeller wet. I still see people start motors with no water. The thought is the power head does not get hot. The problem is they bake the little rubber impeller.
So back to your question it could take what feels like a long time. If your nervous take the cowl off and keep your hand on the power head. If it gets hot to the touch before you start to see some water you may have a problem.

You might just have some sand plugging the small outlet or maybe even a mud dauber/spider plugged it up so don't use the bypass as the end all/be all of your cooling system. If you suspect the engine is cooling (head is cool / water pouring out of the exhaust) blow out that water line (going from the block to the outlet) or stick a small wire in the outlet to ream out whatever is blocking the waterflow.

What motor? It depends on the brand as some have the tell-tale at the beginning of the cooling path and others at the end.
A few days ago I started a Mercury on a Montauk with muffs and it took a very long time for the cooling water to reach the tell-tale. That is normal for a Mercury.
